For Electrophoresis Buffer
10X
TRIS/BORATE/EDTA (TBE)(replaces 50XTAE)
Dry
Ingredients:
- 1g NaOH
MW 40g
- 108g Tris
Base (AKA Trizma Base) MW 121.10g
- 55g Boric
Acid MW 61.83
- 7.4g EDTA
(disodium salt) MW 372.24
Add
dry ingredients to 700mls deionized or distilled water in 2 liter
flask (water should be stirring for best results) Stir to dissolve.
Add
deionized or distilled water to bring up total volume to 1 liter.
Helpful
Hints :
Can
store at room temperature indefinitely (just make sure to avoid
evaporation--you can put it in the refrigerator to help).
Can
reuse buffer
Use
gentle heat to dissolve precipitates that form during storage.
This
makes 1L of 10x so to dilute it to 1X (which is what you'll use
for your labs) you will use 1 ml of 10x TBE for every 9mls deionized
or distilled water. So, to mix up 1L of 1X TBE you would add 100mls
of 10X TBE to 900mls deionized or distilled water.
